陆士新课题组主要从事肿瘤病因与癌变机理的研究,证明了食管癌高发区人群暴露于亚硝胺的水平比低发区高,提出了林县食管癌亚硝胺病因,为林县食管癌预防提供了科学基础;从林县食管癌组织中克隆了4个新食管癌相关基因1-4(ECRG1-4,基因文库登记号为:AF071882.1;AF268198;AF325503),他们对ECRG1、ECRG2、ECRG4的结构与功能进行了系统的研究,其研究结果发表了10多篇论文,分别刊登在Int. J Cancer;BBRC;Carcinogenesis等杂志上。
近年来,课题组又进行了不同肿瘤中肿瘤干细胞的分离与鉴定,在鼻咽癌和食管癌细胞中发现了类似肿瘤干细胞(SP细胞),并对发现的肿瘤干细胞的生物特性如:细胞增植、细胞的自我更新和分化进行了研究,观察到SP细胞植入免疫缺陷小鼠体内有很强的致瘤能力,这些结果对致瘤作用和治疗抵抗的机理均展示了光明的前景,这有助于鼻咽癌和食管癌发现有效临床治疗的新靶点。同时用正常干细胞治疗肿瘤的研究已取得初步令人满意的结果;研究microRNA调控肿瘤干细胞的自我更新与分化也在进行中。历年来曾获多项国家与部级奖。
Research Description
The scientists in this group have mainly engaged in cancer etiology and carcinogenesis. They have proved that the exposure of N-nitrosamines in residents from high risk areas for EC was higher than that from low risk areas. They have contributed to establish N-nitrosamine etiology of esophageal cancer and provided the preventive measures and scientific bases for prevention of esophageal cancer in Lin-xian county.
They have cloned and identified 4 novel Esophageal Cancer Related Genes from human esophageal cancer in Lin-xian county. (ECRG 1-4, GenBank Accession No. AF 071882.1, AF 268198, AF 325503) They have systematically studied the structure and function of ECRG-1, ECRG-2 and ECRG-4 genes. More than 10 papers published in Int. J. Cancer, BBRC, Carcinogenesis, e.g..
Recently several years this group has studied the isolation and identification of cancer stem cells in various cancers. They found the Cancer Stem Cell–Like Side Population Cells ( SP cells) in Human Nasopharyngeal Carcinoma Cell (NPC) Line and in Human Esophageal Carcinoma(EC) Cells. They scanned SP cells from cell lines and investigated stem cell characteristics, such as proliferation, self-renewal, and differentiation. They observed a strong tumorigenesis ability of SP cells following in vivo transplantation into non-obese diabetic/severe combined immuno-deficient mice. These data shed light on tumorigenesis and therapeutic-resistant mechanisms, which are helpful for developing novel targets for effective clinical treatment of NPC and EC.
This group searches to therapy the tumors using normal adult stem cell. They got good results. In addition, they also study the microRNA regulates self-renewal and differentiation. of cancer stem cells.This group has got some national and ministerial awards.
