研究方向
认识我国北方家族性食管癌遗传易感性。建立和维护山西阳泉食管癌高发现场,采集食管癌家系的遗传样本,建立表型不一致同胞对(DSP),通过全基因组相关分析(GWAS)探讨单核苷酸多态性(SNP)和拷贝数变异(CNV)与食管癌易感性的关系。探讨临床医学研究中样本采集的科学问题,参与建立遗传样本采集的科学规范。
在分子水平上发现上皮细胞与癌细胞之间的差异分子,特别是血浆和体液中的分泌蛋白。参与国际人类血浆和肝脏蛋白质组研究计划,建立人类血浆和肝脏蛋白质组表达谱。分离和鉴定食管癌、肝癌、胰腺癌等消化道肿瘤标志蛋白/小分子肽(谱)、自身抗体、适配体以及循环肿瘤细胞,认识这些分子在肿瘤发生发展和转归中的作用。
Research Description
Dr. Zhao’s lab mainly focuses on following research areas: firstly, understanding the genetic susceptibility of esophageal squamous cell carcinoma (ESCC) with family history in the north area of China. A high-incident area of ESCC in ShanXi province was established and maintained. The peripheral blood samples of individuals in ESCC pedigrees were collected. Meanwhile, genome-wide association analysis (GWAS) was conducted to identify the susceptible genes and DNA copy number variances (CNVs) based on discordant sibling pairs (DSPs) samples. Secondly, exploring the specimen collection issues in the clinical medical study, and participating in drawing up the standard operation procedures (SOPs) of sampling. Thirdly, identifying the differentially expressed proteins between normal epithelial and tumor cells at the molecular level, especially for the secreted proteins in the serum or body fluid. We had participated in establishment of the reference expression profiles of human plasma and liver tissues in the Human Plasma Proteome Project (HPPP) and Human Liver Proteome Project (HLPP). In addition, candidate diagnostic biomarkers, including proteins, peptides, serum autoantibodies, aptamers and circulating tumor cells (CTCs) for ESCC, hepatocellular carcinoma (HCC) and pancreatic cancer (PC) were identified and validated through proteomic strategies, and the roles of some key proteins in tumorigenesis and progression were further determined.
